The neuroinflammation in Alzheimer's disease (AD) was assumed to show at late stage of AD. However, recent researches revealed that immune activations contribute to and drive AD pathogenic process in pre-clinical stage. These reactions are occurred by CNS-resident immune cells (microglia) and blood-derived innate/adaptive immune cells (monocytes, macrophages, lymphocytes) and trigger the AD. In this study we examined how theses immune cells are structured around pathology such as amyloid plaques, and at which point they are activated. The brains of 5XFAD AD mouse model were transparent by the CLARITY method and stained amyloid and immune cells including astrocytes marker, GFAP. These images were reconstructed to 3D images of the mouse brain. These 3D images revealed the structural information about migration and activation of microglia and astrocytes surrounding amyloid plaques. And these structural information provides information 3D distribution of immune cells around amyloid deposits and the clues of the role of immune cells in the AD.
