Neurons can become immunological targets of cytotoxic CD8+ T cells in the context of autoimmune or virus-induced encephalitis that are associated with neuronal degeneration and synaptic loss. However, the sequence of cellular and molecular events causing synaptic alterations in these conditions remains to be elucidated. Exploring a murine model of viral infection as well as human autoimmune encephalitis, the presentation will shed light into disease relevant pathways that results in synaptic engulfment by bone marrow-derived and brain-resident phagocytes as a consequence of a pinpointed attack by MHC-class I restricted T cells. The central importance of neuronal signaling pathways enforcing a tripartite interaction between T cells, neurons and phagocytes will be illustrated.
